Detection of sperm antigens on mouse ova and early embryos☆
[摘要] Serumandcolostrumantibodiesagainstmousespermweredevelopedintworabbitsaftersystemicandmammaryglandimmunizations.Indirectimmunofluorescenceutilizingfluorescein-labeledgoatantiseraagainstrabbitIgGandIgA,respectively,indicatedthatbothimmuneserum(IS)andcolostrum(IC)comparedwithcontrolsamplescausedintensivestainingoftheacrosomeandtailofsperm.AbsorptionofISandICwithmouseserumandthespleen,kidney,liver,andbrainofmalemicedidnotreducethestrengthorthepatternofstainingreactiononsperm.TheabsorbedISreactedwithcellsurfacesofoocytes,unfertilizedova,zygotes,two-cellandfour-toeight-cellfertilizedova,andblastocysts.TheabsorbedIC,however,reactedonlywiththefour-toeight-cellembryoandblastocyst.FurtherabsorptionoftheISwithmouseovaryremovedthereactionwithunfertilizedovaandtheone-totwo-cellfertilizedova,butthestainingoflaterembryostageswasunaffected.Therefore,itappearsthatspecificrabbitanti-spermantibodiesaredetectingtwocell-membraneantigensonmouseembryos:oneoriginatingfromtheovaryandtheotherarisingafterfertilization.
