A biochemical study of the effects of cAMP pulses on aggregateless mutants of Dictyostelium discoideum☆
[摘要] Wehaveexaminedtheabilityofsixaggregatelessmutantstoexpressthebiochemicalparametersinvolvedintheproductionofthechemotacticsignal,cAMPpulses.ThemutantsarerepresentativeofthethreegroupswhichhavebeendefinedpreviouslyintermsoftheabilityofappliedcAMPpulsestoinduceall,orpart,ofthedevelopmentalcycleofnonaggregatingstrains[Darmon,M.,Brachet,P.,andPereiradaSilva,L.(1975).Proc.Nat.Acad.Sci.USA72,3163–3166;Darmonetal.,1977].Strainsweremonitoredforadenylatecyclase,cellularandextracellularphosphodiesteraseactivities,thelevelsofcAMPbindingsites,andtheextracellularphosphodiesteraseinhibitor.TheabilityofappliedcAMPpulsestoalteranyoftheseparameterswasalsoexamined.TheresultsindicatethatadenylatecyclasecanexistinactiveandinactiveformsandthatappliedcAMPpulseseffectthetransitionoftheenzymetoitsactivestate.AppliedcAMPpulsesarealsoshowntoinducetheadenylatecyclaseactivityoftwostrainsandinhibittheproductionofthephosphodiesteraseinhibitor.Thedataarediscussedintermsofthepossiblesequenceofeventsinvolvedinthedifferentiationofcellstoaggregationcompetence.
