Effects of amino acids and glucose on adenylate cyclase and cell differentiation of Dictyostelium discoideum☆
[摘要] StarvationtriggersthedifferentiationofDictyosteliumdiscoideumamoebastoaggregationcompetence.Todeterminemorepreciselythenatureofthestarvationsignal,theabilityofvariouscomponentsofthegrowthmediumtoinhibitdifferentiationwasexamined.Changesinadenylatecyclase(theenzymewhichgeneratesthecAMPpulsesbasictothedifferentiationprocess),variousphysiologicalandbiochemicalmarkersofdevelopingcells,andtheabilityofamoebastoformspecificintercellularcontactsweremonitored.Weshowthataminoacidmixturesinhibitcelldifferentiationbypreventingtheincreaseofadenylatecyclaseactivitywhichnormallyoccursduringtheearlyhoursofstarvation.Highconcentrationsofglucosealsoinhibitthedifferentiationprocessbutatalaterstage:Theriseinadenylatecyclasestilloccurswhencellsarestarvedinthepresenceofsugar,buttheenzymedoesnotappeartofunctioninvivo.ExogenouslygeneratedcAMPpulsesarenotabletobypasstheblockexertedbyaminoacidsbutcanbypasstheblockexertedbyglucose.Resultssupportthehypothesisthatthepresenceofaminoacidsinhibitsadenylatecyclasesynthesis,whilethepresenceof3%glucoseblocksendogenousactivationofadenylatecyclase,perhapsasaconsequenceofhighosmoticpressure.
