Spontaneous frequency of a developmental mutant in Volvox☆
[摘要] Twotypesofmutants,thoseresistanttothebaseanalog5-bromo-2′-deoxyuridine(BrdU)andsomaticregenerator(SR)mutants,havebeenanalyzedinVolvoxcarteri.Insomaticregeneratormutants,thesomaticcellswhicharenormallyterminallydifferentiateddedifferentiateandregenerategonidia[Sessoms,A.,andHuskey,R.J.(1973).Proc.Nat.Acad.Sci.USA70,1335–1338;Starr,R.C.(1970).Develop.Biol.Suppl.4,59–100].TheSRphenotypeallowsrecoveryofSRmutationsarisinginsomaticcells,sincesuchsomaticcellswouldregenerategonidiaandgiverisetomutantclones.MutantsofanyphenotypeotherthanSRcanonlyberecoveredifthemutationfirstappearsinagonidium.Sincethesomaticcellsare100-foldmorenumerousthanreproductivecells(gonidia),wehavedeterminedthespontaneousfrequencyofbothsomaticregeneratormutantsandmutationstoBrdUresistanceinordertodetermineiftheSRmutationexertsitseffectinthegonidiumorinthesomaticcell.Thetwofrequencieswerefoundtobenearlyidentical,suggestingthattheSRmutationmustfirstappearinagonidiuminordertobeexpressed.
