The control of avian cephalic neural crest cytodifferentiation: II. Neural tissues
[摘要] Aseriesofneuralcresttransplantationshasbeenperformedto(1)analyzewhetheravianpremigratorycranialneuralcrestcellsarepluripotentialorrestrictedtospecificdevelopmentalpathwaysand(2)examinetheabilityoftrunkneuralcrestcellstodevelopinanenvironmentusuallyoccupiedbycranialcrestcells.Quailembryos,thecellsofwhichhaveauniquenuclearmarker,wereusedasdonorsandchickembryosashosts.Hindbraincrestcellsgraftedintheplaceofdiencephaliccrestcellsfailedtoformneuronsinallbutonecase,inwhichasmallectopicganglionwasfound.Inthereciprocaltransplants,neuralcrestcellsemigratingfromasegmentofforebraincresttissuegraftedintheplaceofmetencephaliccrestcellsproducedtrigeminalandciliarygangliawhichwerecompletelynormal.Thus,crestcellswhichnormallyneverformganglionicneuronswilldosoifplacedinasuitableneurogenicenvironment.Theseresultsprovethatpremigratoryaviancranialcrestcellsarenotrestrictedtospecificdevelopmentalpathways,butareinitiallypluripotential.Trunkcrestcellsgraftedintheplaceofmetencephaliccrestcellsformneuronalgangliaalongtheproximaltrigeminalmotorrootsbutdonotformnormaltrigeminalganglia.Theserootgangliadonotdisplaynormalperipheralprojections,andplacodecells,anormalcomponentofthetrigeminalganglion,formgangliainectopiclocations.Thus,whiletrunkcrestcellsrespondtothemetencephalicenvironmentandformneurons,theirresponseisdifferentfromthatofcranialcrestcellsinthesamelocation.Whetherthisisduetodifferencesindevelopmentalpotentialorininitialpopulationsizeisnotknown.
