Analysis of glycosaminoglycans within the extracellular environments encountered by migrating neural crest cells☆
[摘要] Thevertebratetrunkneuralcrestoriginatesasanundifferentiatedcellpopulationwhichsubsequentlydifferentiatesinresponsetoenvironmentalcuesencounteredduringitsmigration.Glycosaminoglycansaremajorcomponentsoftheembryonicextracellularenvironmentwhichhavebeenimplicatedasimportantinthecontrolofseveralmorphogeneticevents.Toexaminethehypothesisthatglycosaminoglycansmayalsoaffectthedevelopmentoftrunkneuralcrestderivatives,glycosaminoglycanswithinrelevantembryonicregionswerecharacterizedbymicro-spectrophotometricmeasurementsofenzyme-sensitivealcianbluestaining.Theresultsindicatethatthetrunkneuralcrestinitiatesmigrationinanextracellularmatrixrelativelyandabsolutelyrichinhyaluronicacid.Bythecompletionofcrestcellmigration,themajorderivativesofthetrunkneuralcrestaresurroundedbyextracellularenvironmentscontainingdifferentandcharacteristicconcentrationsofhyaluronicacidandchondroitinsulfate.Thepresumptivemelanoblastsandsympathoblasts(whichsubsequentlyundergoasecondarymigration)arebothfoundinregionsofmesenchymewhichcontainhigh,althoughdifferent,levelsofextracellularglycosaminoglycans.Incontrast,thenascentspinalganglioncontainsrelativelylittleextracellularglycosaminoglycan.Theseresultssuggestthatglycosaminoglycansmaybeimportantinthedifferentiationofthetrunkneuralcrest.
