Translation of maternal messenger ribonucleoprotein particles from sea urchin in a cell-free system from unfertilized eggs and product analysis☆
[摘要] MaternalmRNPparticleswereisolatedfromthepostribosomalsupernatantfluidofunfertilizedseaurchineggs.Theyweretranslatedinacell-freesystemderivedfromunfertilizedeggs.Thetranslationoftheseparticlesrequiredthepresenceof12mMMgCl2,whichisconsideredveryhigh.ThesamehighMg2+requirementwasobservedwhenmRNPparticlesweretranslatedinacell-freesystemfrommorulaembryos.Incontrast,mRNAextractedfrommRNPparticlesistranslatedat3mMMgCl2.ThisconcentrationofMg2+isknowntobeoptimalforinitiationofmRNAtranslation.Likewise,arabbitglobinmRNAisfaithfullytranslatedintoαandβglobinchainsinacell-freesystemfromeggsat3,butnotat12,mMMgCl2.ThetranslationalproductsdirectedbymRNPorbymRNAderivedfrommRNPwereexaminedintwogelsystemsandwerefoundtobeverysimilar.Inbothcases,histoneswereidentifiedaspartofthetranslationalproduct.ThisindicatedthatthetranslationofmRNPinhighMg2+isnotduetononspecificbindingoftheseparticlestoribosomes.Theratesofglobinsynthesisinacell-freesystemderivedfromeggsiscomparabletothatofmorularibosomesandtothatreportedfortranslationofglobinwithmouseliverandreticulocyteribosomes,indicatingthatunfertilizedseaurchineggribosomesdonotpossessatranslationalinhibitorandthatnodeficiencyininitiationfactorsformRNAtranslationcouldexplainthelowrateofproteinsynthesisinunfertilizedseaurchineggs.
