Differentiation and competence of cells from hybrid embryos☆
[摘要] CellsfromthelethalhybridgastrulaRanapipiens♀×Ranasylvatica♂wereculturedinvitroassmallaggregates.Thesecellssurviveandundergoextensivedifferentiation,whereasleftwithinthearrestedgastrulatheydie.Incompatibilitybetweennucleusandcytoplasmofthisdiploidhybridappearstoresultinproductionofinhibitorswithintheembryowhichareatleastpartiallylosttothemediumwhensmallcellaggregatesarecultured.Competenceofhybridpresumptiveepidermiscellsforinductionofmelanoblastswasfoundtobelowerquantitativelythancompetenceofnormalcells.Hybridmarginalzonecellsdifferfromnormalschieflyintypeofmusclethatdifferentiates.Functionalcardiacmuscleinhybridssupplantsthevoluntary,striatedmusclecharacteristicofnormals.CellsfromthishybridembryobehavedifferentlyfromnormalRanapipienscellsduringoperationalprocedures,anddifferentclutchesofhybridembryosvaryastoextentofgastrulationandfurtherdevelopmentbeforeonsetofarrest.CellsfromthelethalhybridRanapipiens♀×Ranaclamitans♂wereunabletosurviveinculture.Theblocktodevelopmentatacellularlevelinthiscrossismanifestevenasearlyaslatecleavage.Cellsfrompigmentpatternmutantswereexaminedfortheircompetencetorespondtoaninductorofmelanocytes.BothRanaburnsiandRanakandiyohipresumptiveepidermiscellscanbeinducedtoformextensiveculturesofpigmentcells.Cellsfromthedoubledominant,however,arelessresponsivetotheinductor.Theuseofhybridembryostostudytheintracellularbasisforcompetenceisdiscussed.
