The effect of cell population density on the developmental fate of reaggregating mouse limb bud mesenchyme☆
[摘要] Eleven-dayembryonicC3Hmouselimbbudsweredissociatedandthecellsofthemesenchymewerepermittedtoreaggregateatcontrolledpopulationdensitiesoncelluloseesterfiltermembranesofuniformsurfacearea(poresize0.45μ).Theresultssuggestedthatalargeproportionofcellswerecapableofaggregatingbothascompactclustersandasmonolayers,therelativeproportionsofeachdependingoninitialcellpopulationdensityonthemembrane.Population-dependentaggregativepatternsarisingearlyinculturedeterminedthenatureofsubsequenthistodifferentiationandmorphogenesis.Cartilagedevelopedincellclusterswhich,havingundergonefusion,wereabletoretaincompactcores.Myogenesisoccurredonthesurfaceofmonolayersheets.Itissuggestedthatattheembryonicstagejustpriortoskeletalcondensationinthelimbbud,thegreatbulkofmesenchymalcellsmaybeintrinsicallycapableofdifferentiatingintomorethanoneconnectivetissuetype.Notwithstandinganycovertdifferentiationthatmayhavetakenplace,theirfateisdeterminedbythetypeofcellularassociationsestablishedinvitro.
