Localization of glucosamine-incorporating materials at epithelial surfaces during salivary epithelio-mesenchymal interaction in vitro☆
[摘要] Submandibularsalivaryepitheliumofthe13-daymouseembryo,followingtrypsin-pancreatinseparationfromitsinvestingmesenchyme,incorporatestritiatedglucosaminewhenincubatedinlabeledtissueculturemediumfor1hourat37°C.Afterbeingculturedfor24hourstrans-filterfromunlabeledmesenchymeofasimilarage,thelabelismanifestedautoradiographicallyashighgraincountbeyondthebasalsurfaceoftheepithelialcells.Graincountsovertheepithelialcellswerefoundtorisetoamaximumbetween2and6hoursincultureandsubsequentlytofall.Graincountsbeyondthebasalcellsurfacewerefoundtoincreaseduringthefirst24hoursinculture.Thesedataareinterpretedtoindicatethatlabelappearingatthebasalcellsurfaceby24hoursinculturerepresentsanepithelialcellproduct.Suggestiveevidencethatthelabeledmaterialisamucopolysaccharidecomesfromthefactthathyaluronidaseremovesthelabelwhereascollagenasedoesnot.Evidenceispresentedthatsurfacematerialsremovedbytrypsin-pancreatinatthetimeofculture,orafter24hours,areimportantincontrollingtheuptakeoftritiatedglucosamine.
