Gamma crystallins in Wolffian lens regeneration demonstrated by immunofluorescence☆
[摘要] Afractioncontainingγcrystallinswasisolatedfromthenewtlens,andanantiserumagainstthefractionwasprepared.Testswithimmunoelectrophoresisandimmunodiffusionindicatedthatthefractionwasnotcontaminatedwithothercrystallinsandthattheantiserumreactedwithγcrystallins,butnotwithothercrystallins.Animmunofluorescentreagentwaspreparedwiththisantiserumandusedtodetectγcrystallins.Inthenormallensthereactionwaspositiveinthefiberareabutnotinthelensepithelium.Withthisimmunofluorescentreagent,appearanceanddistributionofγcrystallinswerestudiedinWolffianlensregenerationofadultTriturusviridescens.Theγ-crystallinreactionwasnegativeintheregeneratesofstagesI–IV,aswellasinthenormalirisepithelium.ThereactionbecamepositiveinasmallnumberofcellslocatedintheinternalwalloflensvesicleatstageV.Subsequently,increasewasobservedinthenumberofpositivecellsandinintensityofimmunofluorescenceineachcell.Allcellswhichcouldberecognizedasprimarylensfibercellswerepositive.LaterthanstageIX,whenthesecondarylensfibercellswereformed,theyallbecomepositive.Inallfibercells,γcrystallinsweredetectedinthenucleusaswellasinthecytoplasm.Noimmunofluorescencewasobservedintheexternallayeroflensvesicleorinthelensepitheliumofthegrowinglens.ItwasconcludedthatinWolffianlensregenerationγcrystallinsbecamedetectableafterthecellsenteredtheterminalcellcycleandwereaccumulatedinthecellsduringlens-fiberdifferentiation.
