Cytoplasmic control of nuclear activity in preimplantation mouse embryos☆
[摘要] Techniqueshavebeendevelopedfortheproductionofheterokaryonsbetween2-and4-cellembryosofmouseandadultsomaticcellsofvariousorigins,usingSendaivirus-mediatedcellfusion.Thesetechniqueswereusedtostudythecytoplasmiccontrolofnuclearactivityin2-and4-cellblastomeres.Itwasfoundthatthecytoplasmof2-cellblastomerescontainedfactorsthatinhibitedRNAsynthesisinpreviouslycompetentmousenuclei.Thesefactorscouldalsoaffecthumannucleiandnucleiderivedfrommalignantcellsinasimilarmanner.Inhibitoryfactorswerelargelylostbythe4-cellstage.AlthoughRNAsynthesisbysomaticnucleiin2-cellcytoplasmwasrepressed,itwasfoundthatthetransportofRNAfromsomaticnucleiwasnot.Thecytoplasmof2-cellblastomereswasfoundtocontaincomponentswhichactivatedRNAsynthesisbyhenerythrocytenucleiwhichnormallysynthesizenoRNA.Thecytoplasmof2-cellembryoswasalsofoundtocontainfactorsthatstimulatedDNAsynthesisinnucleiderivedfromacontact-inhibitedculturewhichnormallysynthesizesnoDNA.
