[摘要] Duringanemicepisodes,goatsandcertainsheepreplacehemoglobinA(HbA=α2β2A)withhemoglobinC(HbC=α2β2C).RabbitserumdirectedagainsteitherpurifiedsheepHbAorpurifiedsheepHbCwasprepared.Bothtypeswereusedtotestwhetherthetwohemoglobinsarefoundinthesamecellduringswitchingbyanindirectfluorescentantibodyassay.UnabsorbedantisheepHbAcross-reactedextensivelywithgoatHbAbuttoalesserextentwithgoatorsheepHbC.Similarly,unabsorbedantisheepHbCreactedwiththeseantigensintheorder:SheepHbC>goatHbC>sheepHbA>goatHbA.Cross-absorptionresultedinseraspecificeitherforsheepandgoatHbAorforsheepandgoatHbC.Thespecificitieswereconfirmedbyindirectfluorescentantibodystainingofsheepandgoaterythrocytescontainingeitheratleast99%HbAoratleast99%HbC.SmearsoferythrocytesfromsheepandgoatsintheprocessofswitchingwerereactedwithoneoftheabsorbedserathenwithfluoresceinconjugatedantirabbitimmunoglobulinG.Thesumofthefractionsstainedbothbyanti-HbAandbyanti-HbCexceeded100%duringtheswitch.MoststrikinglywhenHbAwasreplacingHbC,nearlyallcellsstainedforHbCwhilemorethanhalfstainedforHbA.Thus,thetwohemoglobinsarefoundinthesamecellduringswitching.
