The avoidance of stimulatory artifacts in cell cycle determinations in vitro☆
[摘要] MyogenicculturesexhibitapredictablepatternofgrowthcharacterizedbyaninitiallyexponentialaccumulationofDNA,whichdeclinestoaplateaulevelafterfusion.Thispatternisrelatedtocell-mediatedmodificationsoftheculturemediumandcanbealteredbydeviationsfromtheroutinefeedingschedule.ThedatademonstratethatfreshmediumstimulatesDNAaccumulationinoldercultureswhilethemediumremovedfromsucholder,fusingculturesreducestherateofDNAaccumulationinyoungerculturesandalsopromotesfusion.Underexperimentalconditionswhichrequiresomereplacementofmedium(e.g.,pulse-chaselabeling),sucheffectsarepotentialsourcesofexperimentalartifact.Suchartifactscanbeeliminated,however,bytheproperchoiceofmedium.Theleastperturbationoftheculturesoccurswhenmediumwithdrawnfromsisterculturesofthesameageisemployed.
