Freeze-cleave demonstration of gap junctions between skeletal myogenic cells in vivo☆
[摘要] Developingmusclemassesfromhindlimbsof19-dayfetalratswerefreeze-cleaved,platinumandcarbonreplicated,andexaminedelectronmicroscopically.Gapjunctionswereobservedlinkingcellpairsclearlyidentifiedasmyogenicbythepresenceofeasilyrecognizedandcharacteristicarraysofcrossorlongitudinallyfracturedmyofibrils.Occasionallygapjunctionswerealsoobservedbetweenidentifiednonmyogeniccells,butnonewereobservedbetweenmyogenic-non-myogeniccellpairs.Becausetherecentlyformedconjointmyogeniccellswerealreadyencapsulatedbydevelopingbasallaminaeandnormallywouldhavefusedtoformdiscretemyofibers,wesuggestthatthisreportprovidesadditionalevidencethatgapjunctionsnormallyformimmediatelybeforeandthusperhapsmediatetheinitialeventsofmyogeniccellfusioninvivoaswellasinvitro.
