Morphogenetic interactions between rotated skin cuffs and underlying stump tissues in regenerating axolotl forelimbs
[摘要] Rotationofskincuffs180°aroundthelongitudinalaxisoftheunderlyingtissuesintheaxolotlforelimbresultsinahighpercentageofmultipleregeneratesafteramputationthroughtherotatedskin.Similarresultsoccurafterrotationofonlytheanteroposterior(A-P)axisoftheskin.Rotationofonlytheproximodistal(Pr-Ds)axisoftheskinresultsinnormalregenerateswhereasdorsoventral(D-V)axialskinrotationresultsinsingleregenerateswithsomedisturbancesinsymmetry.RotationofanteriororposteriorhalfcuffsofskinproducesresultssimilartothoseobtainedafterA-Protationoffullskincuffs,androtationofdorsalorventralskinhalvesduplicatestheresultsobtainedbyrotatingfullskincuffsabouttheD-Vaxis.Skincuffsrotatedforperiodsfrom6monthstoover2yearsbeforeamputationarealsocapableofcausingmultipleregeneratestoform.Nosignificantdifferenceinthepercentageofmultipleregenerateswasseenafterskinrotationandlimbamputationthroughshoulder,upperarm,andforearmlevels.X-Radiation(4000r)ofeithertheskinorunderlyingtissuesbeforeskinrotationresultedinsingleregeneratesafteramputation.Ifastripofnormalskinwasturnedperpendicularlytothelongaxisoftheirradiatedunderlyingstumptissues,theregenerativeresponsewasblocked.Insomeoftheaboveexperiments,regenerateswithlongitudinallyduplicatedupperarmandforearmsegmentsappeared.Itispostulatedthatnormallyboththeskinandtheunderlyinglimbtissuescaninfluencemorphogenesisduringregenerationandthattheyworkinharmony.Incontrast,rotatedskinandtheunderlyingtissueseachexertamorphogeneticinfluenceupontheregeneratinglimb,andtheregenerateisnotabletointegratethesedisharmoniousinfluences.Thisisreflectedinthehighlyabnormalmorphologyoftheregenerates.Thenatureofthemorphogeneticinfluencedisruptedbyskinrotationisnotyetknown.
