Incorporation of tritiated adenosine into mouse ovum RNA☆
[摘要] ThetotalRNAofovulatedmouseovahasbeenexaminedbypolyacrylamidegelelectrophoresis.TheamountofRNApresentinthetwomainpeaksobserved,28Sand18SribosomalRNA,hasbeenestimatedas0.20ng.TheRNAofovulatedmouseovawaslabeledbyexposureofgrowingmouseoocytestoadenosine-8-3Hinvivo.Forthispurposeasmallvolumeofaconcentratedsolutionoftheprecursorwasinjectedintotheovarianbursa,andovawerecollectedbysuperovulationatvarioussubsequenttimes.Themajorgrowthphaseoftheoocyteisknowntoliebetween20and6daysbeforeovulation.SignificantincorporationintoeggRNAwasobservedwhenbursalinjectionwasperformedbetween19and7days,butnotbetween5daysand1daybeforeovulation.ThetypesoflabeledRNAinovaovulatedatfiveintervalsbetween19and7daysafterbursalinjectionofadenosine-8-3Horuridine-5,6-3Hwereanalyzedbypolyacrylamidegelelectrophoresis.ThedistributionoflabelonthegelsdemonstratedthatthebulkofthelabelappearedinribosomalRNAandtransferRNA.InadditionlabeledheterogeneousRNAwasestimatedtorepresent10–15%ofthetotalincorporation.
