Preferential synthesis of actin during early development of the slime mold Dictyostelium discoideum☆
[摘要] ThechangesinproteinsynthesisduringdifferentiationofthecellularslimemoldDictyosteliumwerestudiedbySDS-polyacrylamidegelelectrophoresis.Totalcellproteinwasanalyzedfollowinga2-hrpulse-label.Itwasfoundthatduringthepreaggregationstage,comprisingthefirstthirdofthedevelopmentalcycle,asinglemajorbandaccountsformorethan20%ofthetotallabeledproteinonthegel.Thisspecieswasproducedinatleast5–10-foldloweramounts,relativetototalcellproteinsynthesis,invegetativecellsandinlaterdevelopingstages.Actinwaspurifiedfromvegetativecellsandwasfoundtocorrespondtothemajorbandinseveralrespects.ThediscoveryofasingleproteinbeingsynthesizedinsuchquantityataspecificdevelopmentalstageprovidesapowerfultoolfortheisolationofaspecificmessengerRNAmoleculeandforanintensivestudyofallthefactorsinvolvedinregulatingproteinsynthesisinaeukaryoticorganism.
