Synthesis of rabbit globin in a cell-free protein synthesis system utilizing sea urchin egg and zygote ribosomes☆
[摘要] ReportsofthereducedabilityofseaurchineggribosomestoparticipateinsyntheticmRNA-directedproteinsynthesishavefosteredthesuggestionthatthelowproteinsynthesisrateofeggsisduetoribosome-associatedinhibitors.Totestthishypothesiswithanaturalmessage,wehaveisolated80SribosomesandmicrosomalribosomesofseaurchineggsandzygotesandcomparedtheiractivityatsynthesizingproteinfromrabbitαandβglobinmRNAinaKrebsIIascitestumorcell-freesystem.Botheggandzygote80SribosomesrespondedtoaddedmRNAandwereshowntosynthesizecompleteαandβglobinchainsbyCM-cellulosechromatography.Inmostcases,theactivityoftheeggribosomeswasincomparableinstanceshigherthanthezygoteribosomes.AttemptstodeterminethecauseofthisdifferenceindicatedthatitwasnotafunctionofK+orMg2+concentration,typeoftRNAused,orribosomalwashproteins.Fromthesestudiesitisapparentthatseaurchineggribosomesarefunctionalatalevelequivalenttoorbetterthanzygoteribosomes,anditappearsthatthelackofproteinsyntheticactivityinunfertilizedeggsisnotduetothepresenceofapopulationofinhibitedribosomes.
