The influence of L-thyroxine on the change in red blood cell type in the axolotl☆☆☆
[摘要] Thesynthesisofnewhemoglobinsduringaxolotldevelopmentsuggestedtheinductionbythyroxineofapopulationoferythrocyteswithadulthemoglobinand/orhormonalmodificationofpreexistingredcellscontaininglarvalhemoglobin.Asassayisdescribedwhichdemonstratestwomorphologicallydistinctpopulationsoferythrocytesduringdevelopment.At100–150daysofdevelopment,thecelltypechangedandasmallpercentageofcellsofintermediatetypewaspresent.Lysatesfromtransitionalpopulationsofcellscontainedbothlarvalandadulthemoglobincomponents,whereaspopulationsbeforeandafterthechangeoverdemonstratedonlylarvaloradultpatternsonpolyacrylamidegels.Autoradiographyof3H-thymidine-labeledcellsgaveevidencethattheproductionofcellsofthelarvaltypeisgraduallyinhibitedratherthancompletelyterminatedduringthetransition.Immersionoflarvaein4.5and9.9×10−8ml-thyroxineprematurelyinducedthecelltypetransition.Inthyroxine-induced76-day-oldlarvae,thecontinualpresenceofthehormonewasrequiredfortheprematurecompletionofthecelltypechangeandforcompletionofanatomicalmetamorphosis.Itwasconcludedthatthelengthoftreatmentwiththehormone(atthelowconcentrationsused)iscriticalindeterminingthenormalcourseofthyroxine-induceddifferentiation.
