The biochemical and ultrastructural demonstration of collagen during early heart development☆
[摘要] Acorrelativeultrastructuralandbiochemicalstudywasmadeofcardiaccollageninthechickembryo,spanningstages9-to11(6to13somites).Analysis(carboxymethylcellulosechromatography,SDSacrylamidegelelectrophoresisandlevelsofprolinehydroxylation)ofcollagensynthesizedinsitupermittedclassificationofthiscollagenastypeI-like(α1:α2=2:1).Correlativeelectronmicroscopyofheartsfixedinsitushowedtheappearanceofstriatedcollagenfibrilsinthecardiacjelly,thuscomplementingthebiochemicalfindings.Althoughtheelectronmicroscopeshowedthepresenceofdevelopingbasallaminaeandlaminalikematerial,synthesisofthetypeofcollagenreportedasuniquetobasallaminaewasnotdetected,andweproposethatthesebasallaminaemaylacktypeIVcollagen.Embryonicstages9-to11aretheearlieststagesinwhichcollagensynthesishasbeendemonstrated,andthisisthefirstreportoftheoccurrenceofcollagenincardiacjellyofearlyhearts.
