Myoglobin synthesis in embryonic cells: Production by both “red” and “white” muscle in cell culture estimated by radioimmunoassay☆
[摘要] Cellculturesofcardiac,pectoral,andthighmuscleofchickembryossynthesizedmyoglobin,asmeasuredbyincorporationofradioactivelysinedetectedbyradioimmunoprecipitation.Liverandskincultures,althoughactiveinproteinsynthesis,failedtodemonstratemyoglobinsynthesis.Puromycininhibitedmyoglobinsynthesisbythecellcultures.Theelectrophoreticcharacteristicsofthemyoglobinantigensynthesizedbythighandpectoralmusclewereidentical.Myglobinsynthesizingprogenitorcellsattachedtoplasticdishesin1hr,butnotcompletelyin0.5hr.Cells,unattachedat0.5hr,wereenrichedinmyoglobinsynthesizingcells.Incorporationoflysine-U-14Cintomyoglobinwasmaximalinconfluentculturesanditsincreaseparalleledtheincreaseofcellfusioninthecultures.Theabilityofpectoral,whitemuscletosynthesizemyoglobininamannerequivalenttothatofcardiactissuewasunexpectedbecauseofitsfailuretosynthesizemyoglobininvivoandmayindicatethatfactorsinthewholeorganismmayregulatetheexpressionofthismusclecell'scapabilities.
