Depression by hyaluronic acid of glycosaminoglycan synthesis by cultured chick embryo chondrocytes☆
[摘要] Theeffectsofhyaluronicacidontheexpressionofdifferentiationbypurepopulationsofculturedchickembryochondrocytesarestudied.Inmediumcontainingserum,hyaluronicacidhaslittleeffectonsulfateincorporation.Intheabsenceofserum,200μg/mlofhyaluronicacidinhibitssulfateincorporationintohyaluronidase-sensitivematerialassociatedwiththecelllayerbyasmuchas50%.Thiseffectisseenby6hrandisprogressive.Hyaluronicacidhasnoeffectonleucineorthymidineincorporationorsulfateuptake,matrixturnover,orcollagensynthesisunderthesesameconditions.Itisshownthathyaluronicacidinhibitschondroitinsulfatesynthesisandtheaccumulationofnewlysynthesizedproteoglycaninthecelllayer.Theseeffectsarenotproducedbyotherglycosaminoglycans,suchaschondroitinsulfate,keratansulfate,orheparansulfate,orbymonosaccharides,butoligosaccharidesderivedfromhyaluronicacidmimictheeffectofhyaluronicacid.Thesedataindicatethatspecificglycosaminoglycans,suchashyaluronicacid,canaffectthesynthesisandaccumulationofotherglycosaminoglycansandshouldbeconsideredaspossibleregulatorsofspecificcellularactivities.
