Repair of a genetically-caused defect in oogenesis in Drosophila melanogaster by transplantation of cytoplasm from wild-type eggs and by injection of pyrimidine nucleosides☆
[摘要] Eggsproducedbyhomozygousmutantrudimentary(r9,154.5)femalesofDrosophilamelanogasterdieduringembryogenesis,apparentlybecausethemutantfemalefailstoincorporatecertainsubstancesintotheeggduringoogenesis.Theseeggscanberescuedbyinjectingthematthepreblastodermstagewithwild-typenucleiandcytoplasmorwild-typecytoplasmalonefromunfertilizedeggs.Somepreblastodermeggsinjectedwith1.5%ofeggvolumeofcytoplasmfromunfertilizedwild-typeeggswereabletocompletebothembryonicandpostembryonicdevelopmentandemergedasadults,whereasnotasingleuninjectedcontroleggwasabletocompleteembryonicdevelopment.Theeggsofrudimentarymotherscanalsoberescuedbyinjectingeacheggattheblastodermstagewith0.01μgofpyrimidinenucleosides.Theresultsdemonstratethatapyrimidinedeficiencyisthecauseofabortiveembryogenesis,andconfirmthepreviousfindingthattherudimentarymutationleadstopyrimidineauxotrophy.
