The role of acid phosphatase and β-glucuronidase in reopening and remodeling of the developing chick esophagus☆☆☆
[摘要] Biochemicalandultrastructuralmethodshavebeenemployedtodeterminetherelationshipofthelysosomalsystemtoreopeningandremodelingofthedevelopingchickesophagus.Biochemicaldataindicateanincreaseinbothacidphosphataseandβ-glucuronidaseactivityduringesophagealorganogenesis.Theappearanceofthesehydrolaseenzymeactivitiesappearstobeindependentlyregulated.Theincreaseinacidhydrolaseactivitiesparallelstheincreaseintotalproteinatatimewhentheesophagusisgreatlyenlarging.Thegreatestactivityofacidphosphatasewaslocalizedintheanterioroccludedsegmentoftheesophagus.Withreformationofthelumen,thedistributionofacidphosphatasebecomesuniformalongtheorgan.Increasesinhydrolaseactivitiescorrelatecloselywithcelldegenerationandthedifferentiationoftheepithelium.AcidphosphataseisidentifiedcytochemicallyintheGolgicomplexandlysosomesofembryonicepithelialcells.Surroundingmesenchymecontainsonlymodestcytochemicalactivity.DiffuseGolgicomplexesandprimarylysosomespredominateduringearlystagesstudied.SecondarylysosomesandcondensedGolgicomplexesarecharacteristicofdifferentiatingepitheliumatlaterstages.Acidphosphataseactivityappearsselectivelylocalizedinvitalepithelialcells,whiledegeneratingandmoribundcells,whoseappearancesareobligatorytothereopeningprocess,failtodemonstrateenzymaticactivity.Thetemporalassociationofincreasedhydrolaseactivityandcytochemicalmaturationofalyosomalsystemsuggestsanactiveinvolvementofthesysteminesophagealorganogenesis.
