Genetic, cytological, and ultrastructural characterization of a temperature-sensitive lethal in Drosophila melanogaster☆
[摘要] GeneticanalysisofastrainofDrosophilamelanogasterrevealedthatarecessivemutation[l(1)ERts]causingtemperature-sensitiveembryoniclethalityislocatedinthedistalregionoftheXchromosomeapproximatelyatmapposition18.At22–25°Cmutantembryosexhibitnormalviability,andalleggsarrestpriortogastrulationiftheyarerearedat29°C.Themutantisbiphasic,exhibitingamaternaleffectwhichisexpressedthroughoutthefirst8hrofdevelopmentaswellasasecondtemperature-sensitiveperiod(TSP)duringthefirst3daysoflarvallife.Larvaeexposedtotherestrictivetemperature(RT)duringthesecondTSPmustalsospendtheremainderoflarvalandpupallifeandthetimeofnormaleclosionatRTtodieasfullydevelopedpupaewhichfailtoeclose.Lightandelectronmicroscopyofarrestedembryosrevealdisturbancesinthedistributionofnuclei,cytoplasm,andyolkandabnormalconfigurationsofroughendoplasmicreticulum.Thecauseofpupaldeathduringthesecondlethalperiodisunknown.
