Direct evidence for transformation of differentiated iris epithelial cells into lens cells☆
[摘要] Althoughitisgenerallyassumedthatthelensregeneratedinthenewteyeaftercompletelentectomyisformedbycellsderivedfromthedorsalirisepithelium,experimentalevidencesofarobtainedforthistransformationdoesnotruleoutparticipationofcellsfromthedorsalirisstroma.WhenthenormaldorsalirisepitheliumofadultNotophthalmus(Triturus)viridescenswasisolatedandculturedinthepresenceoffrogretinalcomplex,newtlenstissuewasproducedin88%ofcultures.Theselenstissueswerepositiveforimmunofluorescenceforlens-fiber-specificgammacrystallinsaswellasfortotallensprotein.Onthebasisofastudyofstromalcellscontaminatingthesamplesofdorsalirisepitheliumandatestforthelens-formingcapacityinvitroofthedorsalirisstromainthepresenceoffrogretinalcomplex,itisconcludedthatlensformationobservedintheaboveexperimentisnotdependentonthecontaminatingstromalcells.Thisimpliesthat,inWolffianlensregeneration,fullydifferentiatedadultcellscompletelywithdrawnfromthecellcyclearetransformedintoanothercelltype.Anadditionalcultureexperimentdemonstratedthatlens-formingcapacityisnotrestrictedtothedorsalhalfoftheirisepithelium,butextendsintoitsventralhalf.
