Regulation of glycerol phosphate dehydrogenase by hydrocortisone in dissociated rat cerebral cell cultures☆☆☆
[摘要] Dissociatedcellsfromfetalratbrainareasweregrownincultureforupto4weeks.Theyexpressedthenervoussystem-specificinductionofcytoplasmicglycerolphosphatedehydrogenase(GPDH;EC1.1.1.8)byhydrocortisone(HC).HCincreasedthetotalandspecificactivitiesofGPDHindissociatedculturesfromallbrainareasexamined—cerebrum,cerebellum,andbrainstem.HC,however,hadnoeffectontheamountoftotalorsolubleproteinoronthespecificactivityofanothersolubleenzyme,lactatedehydrogenase(LDH;EC1.1.1.27).ThefullyinducedlevelofGPDHin12-daycerebralcellcultureswasabouttwofoldgreaterthanthespecificactivityoftheadultratcerebrum.TheinductionofGPDHwasgreaterincerebralculturesgrowninEagle'sBasalMediumthaninseveralothermediatested.TheinducibilityofGPDHovera48-hrperiodincreasedwithtimeincultureuntilamaximumwasreachedaround21days.TheinductionofGPDHbyHCwasinhibitedbycycloheximide,actinomycinD,andcordycepinbutwasunaffectedbycytosinearabinoside.TheseresultsindicatethatproteinandRNA(inparticularmRNA)synthesesarerequiredfortheinductionbutthatDNAsynthesisisnotrequired.
