Developmental change in a myelin-associated glycoprotein: a comparative study in rodents
[摘要] Indevelopingrats,afucose-labeledglycoprotein(mGP)whichiscloselyassociatedwithmyelinhasahigherapparentmolecularweightinimmaturemyelinincomparisonwithmaturemyelin.Inordertoseewhetherthisdevelopmentaldifferenceoccurredinotherspecies,weinvestigateddifferentrodentsofthesuborderMyomorpha.Glycoproteinswerelabeledinvivobyinjectionof[3H]fucoseof[14C]fucose,andpurifiedmyelinfractionswereprepared.ProteinswereextractedwithSDSandseparatedbypolyacrylamidegelelectrophoresis.Usingdouble-labelingtechniques,the3H-and14C-labeledmGPwerecomparedindifferentsamples.WefoundtheshiftintheapparentmolecularweightofmGPduringdevelopmentinrats,gerbils,andhamsters,butwewereunabletodemonstrateitinmiceandprairiedeermice.Intheselatterspecies,themGPofsmallerapparentmolecularweightwasalreadypresentattheearliestagesatwhichitwaspossibletoisolatemyelin.Althoughsomerodentsdonotexhibitthedevelopmentalchange,allshowedacommonfeature:thepresenceofthesamesmallmolecularweightmGPinmaturemyelin.Itmaybethattheglycoproteinofsmallersizeisaprerequisiteforthenormalcompactionandmaturationofmyelin.
