Clonal analysis of vertebrate myogenesis: II. Environmental influences upon human muscle differentiation☆
[摘要] Invitroproceduresforobtainingthedifferentiationofhumanfetalmusclecoloniesweredeveloped,andthesensitivityofclonaldifferentiationtoenvironmentalinfluenceswasexamined.Humanmusclecoloniesarecapableofdifferentiatingintheabsenceofanexogenouscollagensubstrate.Thedependenceofclonaldiffeentiationupontheadditionofchickembryoextracttotheculturemediumisdeterminedbytheserumtypeusedinthemediumandbythesubstrateuponwhichthecoloniesaregrown.Clonaldifferentiationalsodependsuponconditioningofthemediumbythecolonies.Therateofmediumconditioningisaffectedbyclonaldensityandinitialmediumcomposition.Therequiredmediummodificationisnotspeciesspecificsincemediumconditionedbychickmusclecellsalsopermitstheearlydifferentiationofhumanmuscleclones.Bymanipulatingthevariousenvironmentalparametersdescribedaboveithasbeenpossibletodefineanumberofinvitroconditionswhichpermitanormalrateofcellproliferationbutdonotpermitcellfusion.Resultsfromtheseexperimentsarediscussedintermsoftheirdevelopmentalimplications.
