Production of replicable persistent changes in zygote chromosomes of Rana pipiens by injected proteins from adult liver nuclei☆☆☆
[摘要] Aninvestigationwascarriedouttotestthehypothesisthatcellulardifferentiationisbasedupontheacquisitionofspecificmacromoleculesbythechromosomes.VerysmallamountsofvariousproteinfractionsfromadultRanapipienslivercellswereinjectedintozygotesofthesamespecies.Thealbuminfraction,andtoalesserextentthehistones,wheninjectedcauseahighlyreproduciblecessationofcelldivisionandarresteddevelopmentinthelateblastulastage.Nucleifromarrestedblastulaewereseriallytransplantedforsevengenerationswithoutshowinganyrecovery.Itisconcludedthattheeffectisprimarilyonthechromosomesandisreplicatedduringcelldivision.Cytologicalexaminationofblastulaeobtainedfromsuchnucleartransplantationsshowsthepresenceofnormalandabnormalchromosomecomplementsinthesameindividual.Thisindicatesthatatthetimeoftransplantationthenucleiwerenormal,andthattheabnormalitiesoccurredlaterasaconsequenceofthearrest.Theoriginalchromosomalchangesinducedbytheinjectedmacromoleculesmustthereforebebelowthelevelofmicroscopicdetection.
