Observations on the effect of thyroid and parathyroid secretions on explanted mouse radius rudiments

[摘要] Acomparativestudywasmadeconcerningthehistologicaleffectsofthyroxine(2,0.2,and0.02μg/ml)andofparathyroidextract(1IU/ml)onorganculturesof14-to16-day-oldmouseembryoradiusrudiments.Applyinga48-hourcultivationperiod,thyroxinewasfoundtoactonthecartilaginousandbonycomponentsoftherudiments.Thephenomenaactuallyobservedwerequalitativelyidenticaltosomeofthoseoccurringaftertheapplicationofparathyroidextract.Quantitatively,theeffectsofthyroxinewerelesspronounced.Itisconcludedthatinthemouseembryoradiusrudiment,thyroxineisabletoinduceaconcentration-dependent,mildandlimitedparathyroid-likeeffect.Thephenomenaoccurringintheterminalcartilageswereallconcentratedinoraroundtheareaofhypertrophiccells.Theycanbedescribedas:aconictransformation,alossofazurophiliaoftheintercellularsubstance,andadecreaseoftheamountofhypertrophiccells.Asregardsthelatterphenomenon,thyroxineissuggestedtoinfluenceespeciallytherateofproductionofthehypertrophiccells.Concerningthebonetissue,alossoftypicalosteoblastsandofboneformation,especiallyintheareasaroundthecartilaginousepiphyses,becameevident.Applicationofglycine-C14revealedasignificantloweringoftheglycine-bindingcapacityoftheepiphysealpartsoftherudiments.Thefindingsarediscussedinthelightofthecomplicatedsituationarisingifthyroxineisaddedtomediacontainingserumproteinsthatbindthyroxine.
