Chemical dissolution and in vitro reconstruction of sponge cell adhesions: I. Isolation and functional demonstration of the components involved☆
[摘要] Thepurposeofthisworkwastostudythemechanismofcelladhesionusingaggregationofdissociatedspongecellsastheexperimentalsystem.Amethodofchemicaldissociationwasdevelopedformarinespongesusingcoldcalcium-andmagnesium-freeseawater.Theviable,singlecellsresultingfromthisdissociationwereabletoaggregateanddevelopintofunctionalsponges.Rotation-mediatedaggregationwasusedtostudyvariousaspectsoftheaggregationofspongecells.Usingthesetechniques,aspecificrequirementforthedivalentcationscalciumandmagnesiumwasestablishedforspongecelladhesion.Lowtemperaturewasfoundtoinhibitadhesionofchemicallydissociatedcellsevenwhenthedivalentcationswereaddedbacktothecells.Mechanicallydissociatedcellsaggregatedrapidlyatlowtemperatures.Thisdifferencewasshowntobeduetoafactorreleasedintothesupernatantduringchemicaldissociationofspongetissue.Whenthisfactorwasaddedbacktothechemicallydissociatedcellsalongwithdivalentcations,theyadheredrapidlyatlowtemperatures.Thisfactorwasspeciesspecific,causingadhesiononlyofcellsfromthesamespecies.Theseresultswereinterpretedtoindicatethatthespongecelladhesionwascomposedofthreebasiccomponents:thecellsurface,divalentcations,andanintercellularmaterial.Thesecomponentshadbeenseparatedduringchemicaldissociationandwerecapableofspontaneouslyandspeciesspecificallyreassemblingthemselvestorerormanapparentlynormalcelladhesion.Theimplicationsoftheseresultsformodelsofcelladhesionarediscussed.
