The effect of glucose on respiration of the differentiating slime mold
[摘要] TherespirationofthedifferentiatingslimemoldDictyosteliumdiscoideumatdifferentstagesinitslifecyclewasmeasuredwithandwithoutanexogenousenergysource.Itwasfoundthatonadryweightbasis(microlitersoxygenperminutepermilligramdryweight)respirationishighestattheamebastage,declinestoaplateaufromthemigratingpseudoplasmodiumthroughtheculminationstages,anddropsagainatthesorocarpstage.Glucosestimulatesrespirationmaximallyatthesorocarpstage,inspiteofthefactthatthecellsaremaximallypermeabletoglucoseatpreculmination.TheQO2valuesobtainedforsorocarpsinthepresenceofglucoseequaledthoseobtainedforthepseudoplasmodiumandpreculminationstagesintheabsenceofglucose.TheabsoluteoxygenuptakeofagroupofcellsdifferentiatingonagarinaWarburgflaskwasmaximalattheamebastageanddroppedsharplyduringpseudoplasmodiumformation.Alineardecreaseintotaloxygenconsumedwasobservedfromthemigratingpseudoplasmodiumtothefruitstage.Consideredtogether,thedatasuggestthatnosignificantincreaseinrespirationoccursduringtheculminationprocessandthatacriticaldepletionofendogenousenergysourcesoccursduringdevelopmentatculmination.
