Contribution of BDNF-Mediated Inhibition in Patterning Avian Skin Innervation
[摘要] Multiplefactorsareinvolvedinthedevelopmentandregulationofsensoryinnervationinskin.Thefindingswereportheresuggestthatbrain-derivedneurotrophicfactor(BDNF)-mediatedinhibitionmayplayanimportantroleindeterminingthepatternofsensoryinnervationinavianskin.Inbirds,cutaneousinnervationisrestrictedtodermis,whereaxonsformaringofinnervationaroundthebaseofeachfeather.HereweshowthatbothBDNFmessageandproteinaremoreabundantinavianepidermisthandermiswheninnervationisbeingestablished;theBDNFindermisislocalizedtofeatherbuds.Invitro,BDNFcausedgrowthconesofNGF-dependentdorsalrootganglionneuronstocollapse.Similarly,outgrowthofneuritestowardBDNF-secretingfibroblastswasinhibited.TheinhibitoryeffectsofBDNFappeartobemediatedbythelow-affinityp75neurotrophinreceptor,ratherthanatrkreceptor.Thus,thedistributionofBDNFinembryonicavianskinandtheinhibitoryeffectsofBDNFoncutaneousneuritesinvitrosuggestthatBDNFmaybeimportantinrestrictingaxonsfromenteringtheepidermisandthecoreoffeatherbudsduringdevelopmentinvivo.
