α11β1 Integrin Is a Receptor for Interstitial Collagens Involved in Cell Migration and Collagen Reorganization on Mesenchymal Nonmuscle Cells
[摘要] α11β1integrinconstitutesarecentadditiontotheintegrinfamily.Here,wepresentthefirstinvivoanalysisofα11proteinandmRNAdistributionduringhumanembryonicdevelopment.α11proteinandmRNAwerepresentinvariousmesenchymalcellsaroundthecartilageanlageinthedevelopingskeletoninapatternsimilartothatdescribedforthetranscriptionfactorscleraxis.α11wasalsoexpressedbymesenchymalcellsinintervertebraldiscsandinkeratocytesincornea,twositeswithhighlyorganizedcollagennetworks.Neitherα11mRNAnorα11proteincouldbedetectedinmyogeniccellsinhumanembryos.Thedescribedexpressionpatterniscompatiblewithα11β1functioningasareceptorforinterstitialcollagensinvivo.Totestthishypothesisinvitro,full-lengthhumanα11cDNAwasstablytransfectedintothemousesatellitecelllineC2C12,lackingendogenouscollagenreceptors.α11β1mediatedcelladhesiontocollagensIandIV(withapreferenceforcollagenI)andformedfocalcontactsoncollagens.Inaddition,α11β1mediatedcontractionoffibrillarcollagengelsinamannersimilartoα2β1,andsupportedmigrationoncollagenIinresponsetochemotacticstimuli.Ourdatasupportaroleforα11β1asareceptorforinterstitialcollagensonmesenchymallyderivedcellsandsuggestamultifunctionalroleofα11β1intherecognitionandorganizationofinterstitialcollagenmatricesduringdevelopment.
