Hemopoietic spleen colony studies: I. Growth and differentiation☆
[摘要] Thegrowthanddifferentiationofspleencolonies(discretenodulesofhemopoietictissuearisinginthespleensoflethallyirradiatedmicegivenlowdosesofisogenicbonemarrowcellsintravenously)arediscussed.Criteriaforcountingandidentifyingcolonies,adescriptionofeachofthedifferenttypesofcolonies,andtheirrelativeincidence,sizeandlocationareconsidered.Erythroid,neutrophilic,megakaryocyticandeosinophilichemopoieticcellshaveeachbeenfoundintheformofgrossormicroscopicspleencoloniesofsingledifferentiatedcelltype.Eachofthespleencoloniesinitiallyundergoesdifferentiationintoasinglelineofhemopoiesis.Inthespleen,erythroidcoloniesarebyfarthemostnumerous,followedbyneutrophilicandthenbymegakaryocyticcolonies,undifferentiatedcoloniesbeinguncommonandeosinophiliccoloniesrare.Theincidenceofmixedcoloniesincreaseswithtimeascolonieswithasinglelineofdifferentiatedcellsenlargeandundergosecondandthirdlinesofhemopoieticdifferentiation.Colonysizeandcelltypeshowasignificantcorrelation.Mixedcoloniesarethelargestinsize.Nextlargestareerythroidcolonies,thenneutrophiliccolonies.Usuallytoosmalltobegrosslyvisiblearetheundifferentiatedcolonies,eosinophiliccoloniesandmegakaryocyticcolonies,indecreasingorderofsize.Evidenceiscitedfromthisandotherworksuggestingthatthetypicalcolonyformingunitisasinglepluripotentstemcell,whosecommitmenttoalineofdifferentiationisinfluencedbymicroenvironmentalinfluencesaswellassystemichumoralfactors.
