Autoradiographic study of the incorporation of uridine-3H during myogenesis in tissue culture☆
[摘要] Autoradiographicinvestigationofdifferentiatingthighmuscletissueculturesshowedthatafterbriefexposurestoradioactiveuridineorcytosine,theamountoflabelingofnucleiwithinmusclefiberswassignificantlylowerthanthelabelingofnucleiwithinthemononucleatedcells.Thedecreaseintherateoflabelingwhichoccurredattheonsetoffusionofthemyoblastintomultinucleatedfiberscannotbeascribedtogreaterabsorptionoftheβraysbythecytoplasmofdifferentiatedcellsnorisitduetolackofpenetrationofthelabeledprecursorintothecells.ThedifferenceintheincorporationofRNAprecursorsalsodoesnotseemtoarisefromdifferencesinthemitoticactivityofthevariouscelltypes,sinceinhibitionofmitosisbyX-irradiationdidnotabolishthedifferentialrateofincorporationofuridineintoRNA.Theincorporationof3H-leucine,unlikethatofradioactiveuridine,doesnotseemtodecreasefollowingcellfusion.ApplicationofactinomycinDresultedinahigherinhibitionofaminoacidincorporationintomononucleatedcellscomparedwiththatintomultinucleatedfibers.ThesignificanceoftheseresultsinrelationtothepossibleformationofastabletypeofRNAduringdifferentiationisdiscussed.
