The relation of DNA synthesis to RNA synthesis in developing frog embryos☆
[摘要] Frogembryos(gastrulae,neurulae,andtailbuds)werecutintodorsalectoderm-mesodermandendodermregionsandtheisolatedpartswereincubatedwithuridine-14C.Inrelationtothesizeoftheacid-solublepool,theendodermregionssynthesizelesstotalRNAthandodorsalectodermandmesodermcells,whilecytosinearabinosideandLiClinhibitbothtotalRNAandDNAsynthesis.TheendodermcellssynthesizemoreRNAinrelationtothelevelofDNAsynthesisthandothedorsalectoderm-mesodermcells.UnlabeledRNA:DNAratioswerealsohigherfornucleiandchromatinofthebellycellsthanforthedorsalaxialcells.EmbryosinwhichDNAsynthesiswasretardedwithlithiumchlorideorcytosinearabinosidesynthesizedmoreRNAinrelationtotheamountofDNAsynthesisthandidthecontrolembryos.ThissuggeststhatthereislessinhibitionofRNAsynthesis,comparedtothelevelofinhibitionofDNAsynthesis,bycytosinearabinosideandLiCl.LiClandNaCl(0.25%concentrations)causedanincreaseintheleveloflabeledcompoundsintheacid-solublepools.TheresultsarediscussedinrelationtothespeculationthatthefrequencyofmitosiscanaffectthetranscriptionoftheDNAofthegenome,leadingtothesynthesisofqualitativelydifferentmRNAmoleculeswhichaffectthedirectionofcellulardifferentiation.
