Control of lung differentiation in vitro☆☆☆
[摘要] Lungdifferentiationofchimericrecombinationofchickandmousecomponentshasbeenstudiedundervariouscultureconditions.Thecombinationofmouselungepitheliumwithchicklungmesenchymeexhibitsactivemorphogenesis,whereasthereciprocalcombinationshowshardlyanymorphogenesisunderinvitroconditionstested.Ithasbeendemonstratedthatlungmesenchymalhistogenesisisdevelopmentallydependentonlungepithelium,andfurtherthat(1)gutepitheliuminducedsmoothmuscle,vascularelements,andstromalcells,(2)kidneyepitheliuminducedvascularelementsandstromalcells,and(3)bothdorsalspinalcordandsalivaryepitheliumfailedtoinduceanyofthethreehistotypes.LungepithelialbranchinghasbeendemonstratedtobeinducibleacrossaMilliporefilter,butmesenchymalhistogenesiswaslimitedtoconnectivetissuestromaandafewvascularelements.Aftercompletecelldissociation,reconstitutedchicklungmesenchymeindirectcombinationwithmouselungepitheliumdifferentiatedintosmoothmuscle,vascularelements,andstromalcells.Undertransfilterconditions,reconstitutedmesenchymewaspredominantly“pseudo-epithelioid”innature.Aftercompletecelldissociationofchickwholelungrudiments,reconstitutedrudimentsfailedtodifferentiatesmoothmuscle.
