Collagenolytic activity during mammalian wound repair
[摘要] Collagenolyticactivityhasbeendemonstratedinculturesoftissuesfromcutaneousmammalianwoundsculturedonsubstrategelsofreconstitutedcollagen.Asmuchas90–100%oftheinsolublecollagensubstratewasbrokendownatneutralpH,andatleast60%oftheproductwasdialyzable.Inwoundedgeculturescontainingbothepitheliumandmesenchyme,collagenolysiswasinitiallylocalizedaroundtheformer.Variablecollagenaseactivitywasproducedbygranulationtissuealone,butnonebydermisorbyunwoundedlooseconnectivetissue.Unwoundedskinrevealedweakercollagenolyticpotential.Recombinationofseparatedepitheliumandgranulationtissuefromwoundsrestoredcollagenaseactivitytohighlevelsapproximatingthatofunseparatedtissues.Exposureofnonlyticgranulationtissuetoepitheliumappearedtoinducecollagenolyticactivitywhichcontinuedinthemesenchymaltissuefollowingremovaloftheepithelium.Theactivityofisolatedepitheliumdecayedmorerapidlythandidthatofthe“activated”mesenchyme.Viablecellswererequiredforcollagenaseproduction;freeze-thawinginactivatedtheexplants.Enzymeactivitycouldnotbedetectedintissuehomogenatesorextractsandwholeserumapplieddailytoculturesblockedcollagenolysis.CollagenaseactivitywasnotalteredbyvitaminCdeficiency.
