FGF Signaling Regulates Expression of Tbx2, Erm, Pea3, and Pax3 in the Early Nasal Region
[摘要] Fgf8isrequiredfornormaldevelopmentofthenasalregion.Here,wehaveusedacandidateapproachtoidentifygenesthatareinducedinchicknasalmesenchymeinresponsetoFGFsignaling.Usinganexplantculturesystem,weshowthatexpressionofthetranscriptionfactorsTbx2,Erm,Pea3,andPax3,butnotPax7,innasalmesenchymeisregulatedbyectodermalsignalsinastage-dependentmanner.UsingbeadssoakedinrecombinantFGFproteinandanFGFreceptorantagonist,wefurthermoredemonstratethatFGFsignalingisnecessaryandsufficientforexpressionofTbx2,Erm,Pea3,andPax3,buthasnoeffectonPax7expression.Wealsoshowthat,withinthenasalmesenchyme,competencetorespondtoFGFsignalingisinitiallywidespreadanduniformbutbecomesrestrictedtoregionsnormallyexposedtoFGFatlaterstagesofdevelopment,coincidentwithchangesinFGFreceptorexpression.Finally,weprovideevidencethatFGF8alsoregulatesErmandPea3expressioninthenasalplacodes.Together,theseresultsidentifyTbx2,Erm,Pea3,andPax3asdownstreamtargetsofFGFsignalinginthefacialareaandsuggestthatthesegenesmaymediatesomeoftheeffectsofFGF8duringdevelopmentofthenasalregion.
