Regulation of Synaptotagmin Gene Expression during Ascidian Embryogenesis
[摘要] Theascidianembryo,amodelfortheprimitivemodeofchordatedevelopment,rapidlyformsadorsalnervoussystemwhichconsistsofasmallnumberofneurons.Here,wehavecharacterizedthetranscriptionalregulationofanascidiansynaptotagmin(syt)genetoexplorethemolecularmechanismsunderlyingdevelopmentofsynaptictransmission.Insituhybridizationshowedthatsytisexpressedinallneuronsdescribedinpreviousstudiesandtransientlyintheembryonicepidermis.Neuronalexpressionofsytrequiresinductionfromthevegetalsideoftheembryo,whereasepidermalexpressionoccursautonomouslyinisolatedectodermalblastomeres.Introductionofgreenfluorescentproteinreportergeneconstructsintotheascidianembryosindicatesthatagenomicfragmentofthe3.4-kb5′upstreamregioncontainspromoterelementsofsytgene.Deletionanalysisofthepromotersuggeststhatsytexpressioninneuronsandintheembryonicepidermisdependsondistinctcis-regulatoryregions.
