Drosophila APC2 and APC1 Play Overlapping Roles in Wingless Signaling in the Embryo and Imaginal Discs
[摘要] Theregulationofsignaltransductionplaysakeyroleincellfatechoices,anditsdisregulationcontributestooncogenesis.ThisdualityisexemplifiedbythetumorsuppressorAPC.Originallyidentifiedforitsroleincolontumors,APCfamilymembersweresubsequentlyshowntonegativelyregulateWntsignalinginbothdevelopmentanddisease.TheanalysisofthenormalrolesofAPCproteinsiscomplicatedbythepresenceoftwoAPCfamilymembersinfliesandmice.Previousworkdemonstratedthat,insometissues,singlemutationsineachgenehavenoeffect,raisingthequestionofwhetherthereisfunctionaloverlapbetweenthetwoAPCsorwhetherAPC-independentmechanismsofWntregulationexist.WeaddressedthisbyeliminatingthefunctionofbothDrosophilaAPCgenessimultaneously.WefindthatAPC1andAPC2playoverlappingrolesinregulatingWinglesssignalingintheembryonicepidermisandtheimaginaldiscs.Surprisingly,APC1functioninembryosoccursatlevelsofexpressionnearlytoolowtodetect.Further,theoverlappingfunctionsexistdespitestrikingdifferencesintheintracellularlocalizationofthetwoAPCfamilymembers.
