Detection of Neuraminidase Inhibitor Resistant Influenza A/H3N2 Viruses by Nucleotide Sequencing Method
[摘要] BackgroundandObjective:InfluenzaAvirusesarethemajorcauseofepidemicsandpandemicsworldwide.Manyresearchesareunderwayinordertopreventandtreat.RecentlyOseltamivir(Tamiflu),aneuraminidaseinhibitor(NAI),hasbeenusedfortheirtreatmentandprevention.Thepitfallofusingantiviraldrugsisemergenceofresistantmutants.DifferentresearchesinenzymeactivesitemutationanalysisofNAIresistantmutantsrevealedthatcommonmutationsitesarelocatedat119,274,292residues.TheaimofthisstudywasdetectionofR292KandH274YmutationsincirculatinginfluenzaA/H3N2virusesbyusingnucleotidesequencingmethod.MarialsandMethods:InordertoamplifyNAgenemutationsites,weusedconventionalRT-PCRassayon50influenzaA/H3N2isolatesandthenPCRproductsweresequenced.EvaluationandanalysisofrawdatabyBioeditversion7.1.3.0(2011)softwarewasperformed.Results:EvaluationofnucleotidesequencesdidnotshowanyisolateswithR292KandorH274YmutationsandalltheexaminedvirusesweresensitivetoNAI.Conclusion:ThefindingsofthisstudydemonstratethatcirculatingA/H3N2virusesaresensitivetoNAIandlackresistantmutationsites.Asdetectionofdrugresistantmutantswithhighsensitivityandspecificityisveryimportantforefficienttreatmentstrategies,wesuggestnucleotidesequencingasachoicemethodinlaboratoriesprotocolbecauseofitsaccuracy,capabilityandhighefficiencyformonitoringofinfluenzadrugresistantmutantsinpopulation.
