Evaluation of CCND2, PLAB, PCSK2 and FHL1 Genes Expression Efficiency to Differentiate between Papillary Thyroid Carcinoma and Benign Tumors
[摘要] BackgroundandObjective:Fineneedleaspiration(FNA)isthebestmethodforsamplecollectionindiagnosisofthyroidtumorsbeforesurgerywhichisperformedwidelyforpatients,butunfortunatelyforabout20%ofcases,thetestresultisreportedassuspiciousorintermediate.Also,duetofalsepositiveandnegativeresponses,thereisapressingneedforabiomarkertoimprovetheaccuracyofpreoperativetests.TheaimofthisstudywastoevaluatetherelativemRNAexpressionofgenesCCND2,PLAB,PCSK2andFHL1indifferentiatingbenignandmalignanttumorsofpapillarythyroidcarcinoma.SubjectsandMethods:Fiftysamplesfrompatientswithmalignantthyroidtumors(25cases)andbenignthyroidtumors(25cases)wereenrolledinthisstudy,RNAwasextractedfromparaffinsampleswithHighPureRNAParaffinKitandRNAextractedwasusedforcDNAsynthesis.RealTimePCRreactionswereperformedinduplicateconsistsof20lofthereactionmixturecontainingthefluorescentcybergreen.Results:TherelativemRNAexpressionofCCND2,PLABandPCSK2geneswerenotefficientfordiagnosis.However,therelativemRNAexpressionofFHL1genecoulddifferentiate21malignantcaseswith4falsepositiveresults.Conclusions:TherelativemRNAexpressionofFHL1geneismoreefficienttodifferentiatepapillarythyroidcarcinomafrombenigntumors.
