Comparison of Effectiveness and Side Effects of Combination of Memantine and Methylphenidate with Methylphenidate alone in Treatment of Children with Attention-Deficit/Hyperactivity Disorder
[摘要] BackgroundandObjective:Attentiondeficit/Hyperactivitydisorder(ADHD)isacommonchildhoodpsychiatricdisorderanditsearlydiagnosisandtreatmenthavesignificantroleincontrolofsymptomsandpreventionofthesubsequentcomplicationsinchildren.ThisstudyassesseseffectsofcombinationofmemantineandmethylphenidateintreatmentofchildrenwithADHD.SubjectsandMethods:Thisstudywasadouble-blindedclinicaltrial.Thirty-sixchildrenwithADHDweredividedrandomlyintwoequalgroups(18ineachgroup).Thefirstgroupreceivedacombinationofmethylphenidateandmemantineandthesecondgroupreceivedmethylphenidateandplacebo.ThetherapeuticeffectwasassessedbyConner’sscaleandsideeffectsweredeterminedineachgroupatthebeggingandattheendofthefirst,second,thirdandfourthweeksafterintervention.Results:Conner’stestscoresdidnotshowstatisticallysignificantdifference(P=0.835)betweentwogroupsatthebeginningofstudyandattheendofthefirstweek(P=0.136).However,attheendofthesecond,thirdandfourthweeks,thegroupofmethylphenidatewithmemantinehadmoredeclineinConner’stestscores(P=0.012,0.001and0.002,repectivley)comparedtogroupofmethylphenidatewithplacebo.TheoverallresultsshowedthattheConner’sscaledecreasedfrom22.3to10(Plt;0.001)inthemethylphenidateandmemantinecombinationgroupwhichwassignificantlylowerthaninmethylphenidatewithplacebo(decreasedfrom22.4to13.5)(Plt;0.001).Therewasnostatisticallysignificantdifferencesinsideeffectsbetweentwogroups(Pgt;0.05).Conclusion:ThisstudyshowedthatmethylphenidatewithmemantineismoreeffectivethanmethylphenidatealoneforADHDinchildrenandithassimilarsideeffectsprofile.
