Purification of blood coagulation factor V activator from the venom of Iranian Vipera lebetina
[摘要] BackgroundandObjective:ThevenomofmanyViperidaesnakeappeartocontainproteinsthataffectbloodcoagulation.TheaimofthisstudywastoidentifyandisolateabloodcoagulationfactorVactivatorpresentinthevenomfromViperalebetina.MaterialandMethods:FactorVactivatorwaspurifiedfrom200mgofcrudevenombythreestepsofchromatorgraphywhichincluded:gelfiltrationonsephadexG100,IonexchangechromatographyonDEAEandcelluloseandaffinitychromatographyonheparinagarose.TheeffectoffactorVactivatoronhumanfactorVwasstudiedbymeasuringtheamidiolyticactivityoftheproducedthrombinbyactivatedfactorV(Va).Results:TheresultsofSDS–PAGEidentifiedanactivatoroffactorVasasingleproteinbandwithamolecularweightof29KDa.Thiscompoundwasconverted,inthepresenceofcalciumions,toactivefactorVa.ItalsohadarginineesteraseactivitytowardsubstrateBAEE(-benzoylarginineethyester)andaweakamidaseactivityonS-2222(benzoyl-le-glu-Gly-Arg-p-nitroanilide).Conclusion:TheresultsofthisstudyshowedthattheisolatedfactorisaspecificactivatoronfactorV.
