Identification and Determination of Ochratoxcin A concentration in wheat flour factories in Ahvaz city by using HPLC method

[摘要] BackgroundandObjective:OchratoxinA(OTA)isthemostpoisonousofochratoxins,withnephrotoxicityandcarcinogenicitytoxiceffects.ThisstudywascarriedouttoidentifyanddeterminetheconcentrationofOTAinwheatflourproducedbyfourfactoriesinAhvazcityusingHPLCmethod.SubjectsandMethods:InordertoseparationandquantificationofOTAinproducedwheatflourof4factoriesinAhvaz,8times,eachtime,onecompositesamplesfromwheatflourofeachfactorywhichcomprising20grabsampleswastaken.Anadequateofeachsamplemixedwithmethanol-phosphatebuffer,filtered;analiquotofthefilteredwasdilutedandpurifiedbyrunningthroughaimmunoaffinitycolumn.Thecolumnwaswashedwithwater;thenOTAwaselutedbymethanol.Theelutedwasconcentratedtodrynessunderagentlenitrogenstream.Finally,theresiduewasdissolvedinmobilephaseandanalyzedbyHPLCusingcolumn(150mm×4.6mmID,5µmC18stationaryphase)withafluorescencedetector(excitationandemissionwavelengthwas333nm460nmrespectively),mobilephaseofacetonitrile-water-glacialaceticacid(49.5:49.5:1)andatflowrateof1.5mL/minThelimitsofdetectionandquantificationwere0.0075and0.025ng/g,respectively.Results:Thirty(93.75%)outof32sampleswerecontaminatedwithOTA,atconcentrationrangingfrom0.004to0.809ng/g,withanaverageof0.09ng/g.Conclusion:TheresultsshowedthatallofthesampleshadlowerlevelsofOTAthanmaximumtolerablelimitof5ng/gassignedbyInstituteofStandardandIndustrialResearchofIran(ISIRI)forwheatandWHOguidelines.
