Effect of Exercise on Morphine-Induced Toxicity in Rat
[摘要] BackgroundandObjective:Morphineinducesadverseeffectinmanyorgans.Ithasbeenknownthatmoderateexercisereducesoxidativestress.Thepresentstudywasundertakentoevaluatetheeffectofexerciseonmorphine-producedtoxicityinratliverandkidney.SubjectsandMethods:Adultmaleratswererandomlydividedin4groupsincluding:(1)morphine-treated,(2)morphine-treated+exercise,(3)vehicle(normalsaine)treated,(4)vehicletreated+exercise.Animalsingroupsof1and2receivedmorphine(ip)twotimesperdayfor5consecutivedays.1stday5mg/kg,2ndday10mg/kg,3rdday20mg/kg,4thday40mg/kgand5th50mg/kg.Ratsingroupsof3and4receivedvehicleonly.Theanimalsingroupsof2and4wereparticipatedinthetreadmillexercisesessions(1hatspeed17m/min)for10consecutivedays.24hlater,theanimalswerekilledwithoverdoseofsodiumpentobarbital.BloodwascollectedfordeterminationofALT,AST,ALP,BUNandcreatinine.Liverandkidneytissueswereremoved,fixedandprocessedforlightmicroscopy.Tenanimalswereusedforeachgroup.Results:Biochemicalandhistopathologicalobservationsindicatedthatmorphineproducedinjuryintheliverandthekidney.Exercisereducedmorphine-inducedtoxicity(Plt;0.05).Conclusion:Morphine-inducedtoxicityinratliverandkidney.Execiseprotectedliverandkidneyagainstmorphine-inducedtoxicity.Blockingofoxidativedamagebyexercisemaybeausefulstrategyforpreventingmorphineadverseeffects.
